Potency Test of Purified Pertussis Vaccine by ELISA

Abstract
|
|
|
An enzyme-linked immunnsorbent assay(ELISA) was performed for to examine potency test of purified potency vaccine as an alternative method to the current active mouse protective test(AMPT).
For ELISA, purified toxin was used as a coated antigen of ELISA plates. Serum antibody levels against pertussis vaccine of tested animal were measured by Horseradish Peroxidase(HRP) labeled protein G. For AMPT, the protective effect was studied in mice vaccinated with pertussis toxin(PT) according to the WHO recommanded protocol.
There are good relationship of test result´s between ELISA and AMPT(correlation coefficient of r= 0.971).
By ELISA test, test protocol could be simplified and the period required for potency test can be saved by omitting mouse challenge procedure.
